| In vitro study | Sulfacetamide inhibited Arabidopsis DHPS,IC50 = 9.5 μm, pKa = 5.4. Sulfacetamide induces antiproliferative effects in T-47D cells independent of apoptosis and cell cycle arrest. Sulfacetamide decreased the expression of p53/DRAM pathway, and upregulated Akt/mTOR signaling, promoting autophagy. Sulfacetamide has an LD50 of 41 mM in T-47D cells (48 hours of treatment) and does not cause DNA fragmentation. In sulfacetamide-treated cells, ATG5 expression was upregulated, indicating an increase in autophagosome formation. The autophagy response induced by Sulfathiazole and sulfacetamide was not accompanied by apoptosis and cycle arrest. The autophagy response it elicits in T-47D cells does not depend on the DAPK signaling pathway. Sodium sulfacetamide or sulfacetamide is a bacteriostatic agent that is used in Gram-negative and positive bacteria sensitive to sulfonamide (including Streptococci, Staphylococci, E. coli, Klebsiella pneumoniae, Pseudomonas pyocyanea, Salmonella spp., proteus vulgaris, Nocardia) is active. Sulfacetamide inhibits mannose-6-phosphate isomerase(PMI: phosphomannose isomerase), a key enzyme in kinetic energy metabolism. |
| In vivo study | The LD50 of sulfacetamide administered by oral route in mice is 16,500 mg/kg. In humans, side effects include erythema, moderate swelling, nausea, vomiting, and headache. In addition to these side effects, there have been reports of StevensJohnson syndrome in HIV-positive patients using sulfacetamide drops to treat eye infections. However, most of these side effects are caused by the high absorption of the drug through the oral route or through the skin, mucosa and conjunctiva, while topical use of sulfacetamide does not show strong side effects. |
